Donor-specific transfusion in the nude rat prolongs survival of subsequently transplanted hamster cardiac xenografts.

چکیده
XENOGRAFfTUMORS have indefinite survival when transplanted into atbymic nude rats or mice. However, nude rats reject vascularized hamster cardiac xenografts in tempo (3 to 4 days) similar to that of immunocompetent rats.13 This "slow-motion" hyperacute rejection witnessed in the nude rat is thought to be mediated by a T cell-independent, but antibodyand complement-dependent mechanism(s ).1 In this study, we investigated whether prior exposure of nude rat recipients to hamster xenoantigens in the form of donor-specific transfusion (DST), would modulate the humoral response thus allowing for prolonged survival of subsequently transplanted cardiac xenografts.

Pretreatment of recipients (nude rat) with donor antigens leads to prolonged survival of hamster heart xenografts.
THE INITIAL barrier to successful organ xenotrans1. plantation in discordant species is hyperacute rejection (HAR) mediated by either natural (preformed) or induced antibodies. 1•2 Hamster cardiac xenografts are normally rejected by this mechanism within 3 days when transplanted into naive LEW or athymic (nude) rats,3 which in the latter is supposedly caused by a T-cell independent process. Thi...

I T IS KNOWN that donor-specific transfusion (DST) enhances graft survival in clinical transplantation. However, efforts using xenogeneic DST to improve xenograft survival have resulted in hyperacute rejection (HAR) even in the presence of conventional immunosuppression. 1 In this study. we tested the effect of DST combined with FK 506 on survival of hamster-to-rat liver xenografts. متن کاملProlonged survival of rat islet and skin xenografts in mice treated with donor splenocytes and anti-CD154 monoclonal antibody.
Treatment of C57BL/6 mice with one transfusion of BALB/c spleen cells and a brief course of anti-CD154 (anti-CD40 ligand) antibody permits BALB/c islet grafts to survive indefinitely and BALB/c skin grafts to survive for approximately 50 days without further intervention. We now report adaptation of this protocol to the transplantation of islet and skin xenografts. We observed prolonged surviva...
متن کاملDonor species complement after liver xenotransplantation. The mechanism of protection from hyperacute rejection.
Hamster hearts transplanted into stable rat recipients of hamster livers (OLT rats) were hyperacutely rejected after transfer with unaltered rat antihamster hyperimmune serum (HS). This was followed by immediate liver xenograft rejection in 4 of 5 rats. In contrast, simple heat inactivation of the rat HS resulted in prolonged survival of hamster hearts to 25 days without deterioration effect in...
متن کاملLong-term survival of donor-specific pancreatic islet xenografts in fully xenogeneic chimeras (WF rat----B10 mouse).
We recently reported that reconstitution of lethally irradiated B10 mouse recipients with 40 x 10(6) untreated WF rat bone marrow cells resulted in stable fully xenogeneic chimerism (WF rat----B10 mouse). In these animals, the tolerance induced for skin xenografts was highly MHC specific in that donor-specific WF rat skin grafts were significantly prolonged while MHC-disparate third-party xenog...
